Present and Future of Liver Organoid

Professor Dongho Choi gave a talk about present and future liver organoid which is a kind of a specific point of hepatocyte. He started with the information on organoid that had been developed for many years as a steroid and cell mass. Recently, due to a very nice paper from Hans Clever, organoid is defined by multi-cell mass constructed from 3D culture in vitro and it can be generated from various kind of cells. Dr. Choi explained the history of organoid which began from 2009, wherein papers have been published in the literatures from Hans clever about the intestine organoid.

Recently, papers on hepatocyte in on Colonial site papers had been published from various groups. He emphasized that organoid can be regarded as a kind of a system which is an important field of science. For example, the epithelial organoid, multi-tissue organoid, and multi-organ organ is called assembloid. And then when it comes to hepatocyte, there are five different kinds of organoids, such as hepatocyte organoid, horaniocide organoid, pancreas, extrapatic, and intrapatic which is a very well-organized systems that is used in their science. Thinking about hepatocyte organoid as from the Hans Clever's nice paper, liver organoid and hepatocyte organoid are from adult cells but from IPSO embryo stem cell on hepatocyte. The focus on hepatic stem cell is called the chemically derived progenitor derive organoid which is very different because it's from the adult hepatocyte. Dr. Choi made the stem cell from patient hepatocyte and made it from various kind of hepatocyte to stem cell derived organoid.

Professor Choi talked about the chemically derived progenitors of his lab. He has been differentiating various kind of hepatocytes from various current stem cells for many years and one of them is chemically neurotic progenitors. Identification of stem cells from patients happened accidentally as Professor Choi is a surgeon, he got so many livers while doing surgery and he isolated hepatocyte and put some chemicals in the liver and found some chemically derivative progenitors from this for various kind of patients. He then tried to put some chemicals to mouse hepatocyte and human hepatocyte for many years. On the process, some proliferating cells from the hepatocyte of population were found. On the similar period, a Japanese group published a paper about this using YAC and different chemicals from their lab and they treated these three different chemicals and they made a this is stem cells and they differentiated in hepatocyte and biliary cells it was published in the cell stem cell but it was a rodent hepatocyte. A Japanese group failed in human but he succeeds in human as different combination of chemicals to be published in these chemicals in general pathology and he used three of six different patients as a donation and he treated YAC same as Japanese group but he failed to make a human peripherally hepatic stem cell with the YAC treatment with similar data with Japanese group but succeeded in Human by adding HCGF instead of using Y it's called HAC.

The important things about chemical derived organoid is the differentiation of the cells to hepatocyte which is very similar to primary hepatocyte compared to IPS derived hepatocyte or die combat side and are very different from primary hepatocyte. Primary hepatocyte is closer to CDH hepatocyte and stem cell development express plus a metabolic enzyme which can be engraved in various kind of disease model. This kind of stem cells is very good for various kind of regenerative medicine, disease modeling, and field study. Similar data have been published in the Japanese group, Chinese group even Western Group that chemically derived hepatic progenitor is another single event finding that can be believable. Professor also talked about the recent progress of organoid field and concluded that there are several kinds of stem cells in which one of them is chemically derivative projectors. He expressed his viewpoint towards his work and is proud of these stem cells they are focusing on to apply to various kind of standard field. He added that organic research is one of the brand-new technologies which can applied in their center.
